Most living things have circadian rhythms, including animals, plants, and\u00a0microorganisms. In humans, nearly every\u00a0tissue\u00a0and\u00a0organ\u00a0has its own circadian rhythm, and collectively they are tuned to the daily cycle of day and night.<\/p>\n<p>Circadian rhythms influence important functions in the human body, such as:<\/p>\n<ul>\n<li>Sleep patterns<\/li>\n<li>Hormone\u00a0release<\/li>\n<li>Appetite and digestion<\/li>\n<li>Temperature<\/li>\n<\/ul>\n<h3 id=\"what-scientists-know\">What Scientists Know About How Circadian Rhythms Are Controlled<\/h3>\n<p>The system that regulates an organism\u2019s innate sense of time and controls circadian rhythms is called a\u00a0biological clock. It\u2019s composed of\u00a0proteins\u00a0encoded by thousands of\u00a0genes\u00a0that switch on and off in a specific order. A master clock coordinates all the biological clocks in an organism.<\/p>\n<p>In\u00a0vertebrate\u00a0animals, including humans, the master clock exists in the brain. The human master clock is a large group of\u00a0nerve cells that form a structure called the suprachiasmatic nucleus (SCN). Among other functions, the SCN controls production of the hormone melatonin based on the amount of light the eyes receive. In the evening, a person\u2019s master clock tells their brain to make more melatonin, causing sleepiness. The SCN also synchronizes the circadian rhythms in different organs and tissues across the body.<\/p>\n<h3>Health Effects of Disrupted Circadian Rhythms<\/h3>\n<p>Circadian rhythms can fall out of sync with the outside world due to factors in the human body or environment. For example:<\/p>\n<ul>\n<li>Variants\u00a0of certain genes can affect the proteins that control biological clocks.<\/li>\n<li>Neurological diseases, such as\u00a0Alzheimer\u2019s\u00a0<span class=\"extlink-nobreak\">disease<\/span>, can disrupt circadian rhythms, causing poor sleep quality and changes in symptoms from day to night.<\/li>\n<li>Travel between time zones (jet lag) and shift work alters the normal sleep-wake cycle.<\/li>\n<li>Light from electronic devices at night can confuse biological clocks.<\/li>\n<\/ul>\n<p>Drowsiness, poor coordination, and difficulty with learning and focus may occur when circadian rhythms fall out of sync short term. Long-term sleep loss and continually shifting circadian rhythms can increase the risks of\u00a0<span class=\"extlink-nobreak\">obesity<\/span>,\u00a0<span class=\"extlink-nobreak\">diabetes<\/span>,\u00a0mood\u00a0<span class=\"extlink-nobreak\">disorders<\/span>, heart and\u00a0blood\u00a0<span class=\"extlink-nobreak\">pressure<\/span>\u00a0problems, and\u00a0<span class=\"extlink-nobreak\">cancer<\/span>, and can also worsen existing health issues.<\/p>\n<div class=\"row\">\n<div class=\"col-md-8\">\n<h3 id=\"nigms-funded-research\">NIGMS-Funded Research Advancing Our Understanding of Circadian Rhythms<\/h3>\n<p>Researchers are studying circadian rhythms to gain better insight into how they work and how they affect human health. Some of the most pressing questions that scientists seek to answer are:<\/p>\n<ul>\n<li><strong>What molecular mechanisms underlie circadian rhythms?<\/strong>\u00a0Feedback loops that regulate biological clock proteins are an important part of maintaining circadian rhythms. Basic science research aims to identify more of the proteins and\u00a0pathways\u00a0involved in keeping time over 24-hour cycles, responding to external cues such as light and food intake, and synchronizing circadian rhythms throughout the body.<\/li>\n<li><strong>Can scientists develop therapies that target circadian rhythm pathways to treat circadian dysfunction?<\/strong>\u00a0Disruptions in circadian rhythms happen in shift workers, travelers, and patients with\u00a0autism spectrum\u00a0<span class=\"extlink-nobreak\">disorder<\/span>, Alzheimer\u2019s disease, and\u00a0Parkinson\u2019s\u00a0<span class=\"extlink-nobreak\">disease<\/span>, among other conditions. Scientists are looking for therapies that may affect circadian rhythm pathways and help relieve the symptoms of circadian dysfunction.<\/li>\n<li><strong>What genetic variants lead to circadian rhythm dysfunction?<\/strong>\u00a0Some patients have extreme circadian behaviors, including sleep-wake cycles that shift daily. Genetic screens for variants of genes known to be associated with the biological clock can help explain these patients\u2019 symptoms. These screens may also identify genes previously unknown to be associated with the biological clock.<\/li>\n<\/ul>\n<\/div>\n<\/div>\n<div class=\"row\">\n<div class=\"col-md-8\">\n<h3 id=\"research-organisms-used\">Research Organisms Used to Study Circadian Rhythms<\/h3>\n<p>Microorganisms, fruit flies, zebrafish, and mice are often the\u00a0research organisms\u00a0that scientists study because they have similar biological clock genes as humans. For example, the cyanobacterium\u00a0<em>Synechococcus elongatus<\/em>\u00a0has a fully functional circadian rhythm. Using techniques including\u00a0CRISPR\u00a0genome editing, researchers remove clock genes from cells of this cyanobacterium species to shed light on the function of individual proteins. Similar experiments in fruit flies advance the study of the molecular mechanisms underlying circadian rhythms and their effects on behavior. Scientists control their research organism\u2019s environment by altering light and dark periods.
